For various reasons, I keep about half of my music on a drive in my desktop box, and the other half on a usb drive. When I first set up the server software, I pointed it to the folder on my desktop's drive. I assumed that I could then create a link to the music folder on the usb drive in the desktop drive, so that I could access both sets of music. But this does not seem to work. Is there any way to do this?

Shortcuts are the way to go. If it isn't working for you, we'll have to
find out why. Did you do a full rescan after you added the shortcut?

If you turn on d_scan and d_info in Server Settings/Debugging you'll find
information about the scanning process in your log file. This might help
find out what's wrong. Search the log file for the name of your shortcut.
If the file is growing too huge, you might test it with an empty folder as
main music folder in which you put the shortcut to the external disk.
