I have Logitech Media Server on a PC and a laptop (both Windows Vista) to feed music stored on each to a Squeezebox Touch. Just in the last day or so the LMS on the PC has stopped working i.e. SBT can't "see" any music on that PC and when I go into Diagnostics in LMS it comes up with the following error message "Logitech Media Server is running but can't be contacted on port 9000.The following applications might be blocking your connection: -" but doesn't list anything. Helpful.
Have reinstalled LMS but to no avail. All is fine on the laptop and SBT working OK from that. Any ideas?

Port 9000 is a commonly used port for media servers so any number of programs may be trying to access it. Easy fix (assuming that is indeed the issue) is to simply assign a different port # to LMS.
LMS > Settings > Advanced > Network > Web Server Port #
I use port 9010 myself. Click Apply, stop then restart LMS, check diagnostics to see if it has cleared the issue.
Keep in mind if you want to remote access LMS you now must use the new port #.

OK I think I have sussed it. I have recently started using some work software that basically makes my PC look like a PC in the office. This clearly conflicts with LMS as the latter won't work after I exit the work thing unless I reboot, after which all is well. I will riase it with the work IT "people" but won't be holding my breath.

You are very likely using a VPN tunnel a virtual private network an encrypted tunnel to your work network so you are not connected to your own network anymore even if you are electrically but networking can be more than that so this works the same as if you would try to connect while your laptop is at your workplace, in short they are not on the same network laptop and Touch .
Just copy the files to the home computer ?--------------------------------------------------------------------

First, make sure your LMS has a static IP address ! The fact you have to restart LMS after disconnecting may be meaningful.
Many VPN's actually allow dual connections and even without that, it's usually possible to add a couple of static addresses <since XP you can have multiple IP's on the same network adapter.>
